Developement of a half life prolonged Albumin
The topic of the thesis work is to understand the biology behind the long serum half-live of albumin, find ways to modulate the interaction between albumin and its receptor FcRn with the ultimate goal to modify and extend the half-life of a recombinant albumin.
In the thesis the exact binding site of albumin to FcRn will be determined by alanine scanning mutagenesis and in vitro and in vivo tests. Once the amino acids involved in binding will be located, further mutations will be introduced to modify the binding towards a half life extended albumin molecule which could be used for second generation albumin fusion proteins in a variety of indications.
