Abstract

The interferon regulatory factor 8 (IRF8) plays a prominent role during the development of immune cells and their function. Often it loses its functionality throughout a chronic myeloid leukemia due to further mutations. Moreover, a BCR::ABL-independent knockout results in a CML-like disease in mouse models. In contrast, in recent studies oncogenetic properties of IRF8 have been observed, first in AML, later also in CML. In this study, we investigated to what extent IRF8-regulated genes, such as IFIT2 (“interferon induced with tetratricopeptide-motif”), are crucial for this effect. Due to a transgenic mouse model it was possible to generate stem cells bearing a BCR::ABL-translocation, the Irf8-loss as well as a knockdown of Ifit2. Transplanting those stem cells into wildtype recipients resulted in a lethal leukemia, which indicates the importance of IFIT2-functionality for the tumorsupressive effect of an IRF8-Knockout in BCR::ABL-positive stem cells.
