Abstract

TRPC5 becomes sensitive to DAG if 1. the activity of protein kinase C is down regulated or 2. the coupling between the channel and NHERF is disrupted or 3. PIP2 concentration in the plasma membrane is reduced. These findings were made in HEK293 cells over expressing TRPC5 and in HT22 cells which are endogenously expressing TRPC5. Thus DAG appears to be the second messenger to activate TRPC5 after activation of Gq-protein coupling receptors.
