Abstract

Human cytomegalovirus (HCMV) open reading frame UL83 codes for the phosphoprotein pp65 (ppUL83), an abundant constituent of the virion tegument. It is also detectable in DBs and NIEPs. Immediately after infection pp65 is targeted to cell nucleus. This transport is mediated by two nuclear localization signals. New synthesized pp65 is detectable in the cell nucleus 4 hours after infection. Pulse chase experiments were used to show the nuclear transport of pp65 in the late phase of virion replikation. A statement about the quantity of this nuclear transport was not possible. Therefore transfectionexperiments were used to show the nuclear transport of pp65 in vivo. For this a fusionprotein of pp65 and the green fluorescent protein (GFP) was used, which was localized in the pcDNA3.1 vector. Using the confocal laser scanning microscopy, green fluorescent structures were detectable up to 12 hours after infection.
