Individualdifferencesindopamine-relatedtraitsinfluencemoodeffectsofdopamineD2-antagonistandantidepressanttreatmentexpectations

Abstract

Background: Hightraitanhedoniaandlowtraitextraversionhavebothbeenpreviouslyrelatedtonotonlylowstatepositiveaffectbutalsodepressivedisorders,disruptedrewardprocessing,andalteredmesolimbicdopaminergicsignaling.Researchonplaceboresponsessuggeststhattreatmentexpectationsmayalterdopaminesignaling,elevatepositiveaffect,andreducedepressivesymptomsinanhedonicindividuals.However,itremainsunclearwhethersuchantidepressantplaceboresponsesdependonputativelowbaselinedopaminergicfunctioninginhighanhedoniaandlowextraversion.Thepresentstudyinvestigateshowinterindividualdifferencesinthesetraitsinfluencepositiveaffectiveresponsesundermanipulationofdopamineandtreatmentexpectations. Methods: Inarandomized,double-blind2×2design(N=297),weadministeredeitherplaceboorthedopamineD2receptorantagonistsulpiride(400mg),andmanipulatedtreatmentexpectationsbytellingparticipantsthattheyreceivedeitheramood-elevatingdrugoraninactivesubstance.Moreover,weassessedtraitanhedoniaandextraversion,andhadparticipantsratetheirstatepositiveaffectat6differenttimepointsbeforeandaftertreatment. Results: Traitanhedoniaandextraversion,aswellasabroadtraitpositiveaffectivityfactor,predictedstatepositiveaffectacrosstimepoints.Importantly,theeffectsofsulpirideandantidepressanttreatmentexpectationsonpositiveaffectweremoderatedbydopaminergictraitssuchthatsulpirideincreasedstatepositiveaffectinhighanhedoniabutdecreaseditinlowanhedonia.Similarly,antidepressanttreatmentexpectationsraisedpositiveaffectinlowextraversionbutreduceditinhighextraversion. Conclusions: Thisstudydemonstratesthatdopamine-relatedindividualdifferencesmoderatetheeffectsofbothsulpirideandaplacebointerventiononpositiveaffectivestate.
